Öğe Anthelmintic Activity of Nigella sativa against Caenorhabditis elegans(Horizon Research Publishing, 2021) Sen, Emine; Ogut, Tugce; Olgun, Abdullah; Kisa, OzgulIncreasing resistance against classical anthelmintic drugs makes discovering new anthelmintic compounds from natural plants important. Nigella sativa (N. sativa) is used as a medicinal plant overall the world and is known to have anthelmintic activity. Caenorhabditis elegans (C. elegans), a common cost-effective model organism that is easily maintained, is useful to determine the anthelmintic activity of new compounds derived from natural products. In our study we aimed to evaluate through toxicity assays the nematocidal activity of N. sativa on C. elegans during its larval and adult stages. Different concentrations of N. sativa oil (900, 450 and 270 mg/mL) were tested and toxicity assessments were done under stereomicroscope by counting the number of surviving nematodes. This study showed that N. sativa essential oil significantly decreases survival of C. elegans in both larval and adult stages at 900 mg/mL final concentration. Larval-stage worms were more sensitive to N. sativa essential oil than were adults. We recommend further studies on other effects of N. sativa on C. elegans after removing the toxic compound(s) from the extract. The further discovery of N. sativa essential oil compounds responsible for anthelmintic activity and determination of their mechanisms of toxicity can pave the way toward new medicines.Öğe Conference report: The 2nd Symposium of sudden cardiac arrest survival(Turkish Soc Cardiology, 2016) Vural, Mutlu; Olgun, AbdullahUnder the leadership of the heart project, “The 2nd Symposium of Sudden Cardiac Arrest Survival” was held with the cooperation of Hayatta Kal Derneği (Stay Alive Association) and İstanbul Kemerburgaz University Faculty of Pharmacy. The aim of the symposium was to find new approaches to increase the survival rate of out-of-hospital sudden cardiac arrest (SCA), which is unfortunately less than 1% in Turkey. Another purpose was to create a public awareness about the importance of first aid. The symposium was conducted by Mutlu Vural, MD and Abdullah Olgun, MD; both of them were directors and lecturers in the symposium.Öğe "Metformin-resistant" folic acid producing probiotics or folic acid against metformin's adverse effects like diarrhea(Churchill Livingstone, 2017) Olgun, AbdullahMetformin, first line medication in the treatment of type2 diabetes by millions of patients worldwide, causes gastrointestinal adverse effects (i.e. diarrhea) in approximately 30% of patients, frequently leading to discontinuation. Interestingly, metformin was reported to increase life span in a microscopic worm, Caenorhabditis elegans, by decreasing folate and methionine production of bacteria that this worm uses as a food source. Metformin can be expected to have a similar effect on some microorganisms of human gut microbiota. This can disturb the balance of gut microbiota and cause gastrointestinal adverse effects by altering folate production of some types of bacteria and suppress their growth. Metformin resistant probiotics can be discovered or generated by artificial evolution/selection, and used to prevent these adverse effects. Bu nedenle yaşlanmanın mekanizmalarını çözmek muhtemelen çok zor ve zaman alıcı bir süreç olacaktır. Tıbbi bir bakış açısıyla yaşlanma, -pragmatik gerekçelerle bile olsa- herkesi etkileyen bir hastalık olarak kabul edilebilir. Yaşlanmak, gençlerde çok zararlı olmayan bazı faktörlere bağlı ölüm riskini de artırır. Yaşlanmaya yol açan mekanizmalarda doğal seçilimin bir avantajı görünmediği için, yaşlanma seçilmiş özelliklerin bir yan etkisi olabilir. Model organizmalarda yapay seçilimin uzun yaşamı sağlayan yeni mekanizmalar keşfedebilirsek, bunların bazı uygun olanlarını gençleştirici tıpta da kullanabiliriz.
